Good Omens coauthored by Pratchett and Neil Gaiman would be my rec. Standalone book, and awesome.

Alternatively, you can just start reading through Discworld. I suggest reading them in publication order, starting with the Colour of Magic.

I'd have to say Color of Magic would be your best bet. Then go into The Light Fantastic, and I've pretty much skipped around Discworld from there.
